I cant seem to access the start menu ingame i've changed the button around but still can not get it to open up.

Try ALL the buttons - it's most likely not the one you think it should be.

Seems you can read: read your game manual, it should be detailed there which button is used, then look which key is mapped to that button on desmume, and done.
