I downloaded DeSmuMe last night and it was working perfectly for me, but when I went to resume playing this morning, I ran into some difficulty. I can't get the emulator to unpause when I load the ROM.
I have been searching the forums and internet for 2 hours and have been unsuccessful in my efforts to find a solution to my problem. Thank you in advance!

Ok 2 question:
What version of desmume are you using (remember that 0.9.6 is the lastest one)?
What game are you playing? Maybe it's a bug, could be interesting for developer.
Also, have you extracted it into a folder, right?

Version 0.9.6 (64-bit, I checked my computer before downloading if it matters at all)
Pokemon SoulSilver
However, I'm confused. Folder?
Note: I have read the FAQ, but it was working fine last night. I closed the emulator multiple times and was able to resume without any troubles.

Game's got antipiracy measures, making it freeze randomly. We don't support it, but there's plenty of help online.

Try with a "clean" start: delete desmume.ini and reconfigure the emulator on Path settings according to where the game/savegame/state is. That game is well known for random freezes, i suggest you to use ingame save and not SAVESTATES.

@Manivo:
But it isn't frozen. The emulator won't unpause.
I've had the game freeze randomly on other emulators but DeSmuMe is the first one that I have no problems with.
If there isn't anything that I can do, could I please have help converting the to .sav so I can try playing on another emulator? I got about halfway through the game with no trouble and don't want to just start over.

Okay! Thank you.
